To transfer it from one Pokemon to the other, have the infected Pokemon in your party before the uninfected Pokemon. It will transfer it eventually after some walking around to the Pokemon after it. The Pokemon has it when 'PKRS' appears in purple on its Summary page.
After a Pokemon has gone 24 hours with Pokerus, it will go away and not be able to infect other Pokemon again, or get pokerus again. A little face will appear next to the Pokemon on the Summary page to show this. However, the effects of the Pokerus still remain with that Pokemon. To keep Pokerus on your Pokemon, put it in the Box. It will remain infected this way.
