No, the Daily Bugle is not a real newspaper. It was always used in Spider man and other Superhero comics, although the Bugle does have a little bit of a back story to it. The newspaper itself was first came up in a comic issue 2 I think of the Fantastic Four and in Spider man the headquarters were located at Madison Ave. but then was moved to a street in the East Fifties. The daily Bugle was used in the comics and movies as a setting and the papers in the story were used to show what the public knew of the Superhero's fantastic adventure of what he/her did.
