The thing about the letters was that they would have the exact address for Harry, even including the room he was kept in. The first letter had 'cupboard under the stairs' as part of its address line. When Mr. Dursley changes Harry room on seeing this letter, the second letter bears the name of the room where Harry has been changed to. This convinces Mr. Dursley that whoever was sending the letters was keeping watch on the family and knew where Harry was being kept in the house.
