Who is the first villain Spider Man fights?

The Chameleon was his first supervillain. However, before him, he did fight Flash Thompson, even though he did not have powers, he was still very mean to Peter Parker.

-Spider-Man's first brush with action is against a small criminal organization leader named Undertaker, after one of his men tries to con Aunt May. His first run in with a super villain comes in a close encounter with The Kingpin after Spider-Man finds he and an acquaintance named Joey threatening to take a mans life.

The movie actually stayed somewhere true to this part of the comic. It was in Amazing Fantasy #15. This is where Spider-Man made his first appearance. As Peter Parker, his first enemy was Flash Thompson, a school bully. However, shortly after he becomes Spider-Man. He faces off in a wresting match with Crusher Hogan. Crusher isn't so much an enemy as an opponent. After the match, the police were trying to stop a thief, but Peter let him go by with no attempt to stop him. A couple of days later he came home to find the police in front of his home. He found out his uncle had been murdered by the escaped thief. He went to an old warehouse where the murderer was barricaded in and faced off with him. So, in short, Spider-Man's first enemy was his uncle's murderer.
