There are no direct references in the books to Nagini killing a muggle out right. She consumed Charity Burbage after a killing curse was used on her and Nagini was made into a horcrux after her master killed Bertha Jorkins.
No such thing happened in HP4.

Well Voldemort would have 'killed' Harry before he got to attack Nagini because he refused to let Nagini leave his side once he knew Harry was destroying his horcruxes. He would no doubt have also killed Ron, Hermione and then Snape. - although it still wouldn't have worked very well.
