Deadpool is actually one of Major Wade Wilson's many nicknames. Other than Deadpool his other most common nickname is "Merc with a Mouth".

Read the Deadpool Special 'Deadpool and Death' for a more in depth explenation, but basically, when he enrolled in the Weapon X program (Nothing to lose, he had cancer) he became a failed experiment and was sent to a place where he and other failed experiments are experimented on some more except without regard for the lives of the subjects.

The subjects/inmates had a betting pool which they called the 'Dead Pool' in which they betted on who was most likely to die.

Wade Wilson (Deadpool) topped the list with his healing factor, dubbing himself 'Deadpool'.
