Well, loss of sleep will not directly kill you, but problems that arose because of it can kill you.
When you do not get enough sleep, your immune system becomes compromised, your blood pressure can go up, you can develop Diabetes or heart issues also. Your cognitive abilities will be compromised, ability to think straight, make decisions. Driving drowsy is just as bad as driving drunk. People with sleep disorders have been tested in many studies, and their reaction times and decision making were that of a person who was over the legal alcohol limit. So, not sleeping can be very dangerous.
A visit to your doctor, and subsequent appointment with a sleep specialist for a sleep study is the way to go. Good sleep habits, making your room comfortable and quiet, the right amount of exercise not too close to bed time. If you get too much exercise it will bring your core temperature up, and make it harder for you to sleep. Your body needs to cool down to sleep properly. there are many good web sites that cover good sleep habits, etc.
One of those is Web MD. another one is www.sleepfoundation.org
So, yes, lack of good quality sleep CAN kill you in a lot of different ways. It is no joke.
It depends on the speed - At 2 mph it would take five days without sleep - so ten days with sleep.

Sometimes as your body needs to recover from a long day and to take in what has happened through the day.You can get sick by being run down and not getting enough sleep. So definitely make sure you get your 8 hours:)
I recall reading that it would take you 3 weeks to count to a million if you counted all day, taking time off to eat and sleep. So to count to a billion would take 1000 times 3 weeks.
