When a red blood cell matures it pushes out its nucleus out of the cell which changes the shape of the cell.Shape is not cone or doughnut.The shape is entirely different.When nucleus is travelling out of the RBC along with other structures( spectrin,actin, haemoglobin etc. which are coming in the path of the nucleus)creates empty space .This empty space due to its negative pressure becomes flattened .The rest of the area is still thickened which gives a "C" shaped appearence." C " shaped thickened area covers the 2/3rd margin of RBC ( or more ) while the flat area covers the centre of RBC and 1/3rd margin of RBC( It may be less ).The flat central area is concavo-convex while the peripheral flat part is slightly concavo-convex but it is opposite to the central part thus the total flat area is giving an appearance of letter " S ".So the total appearance of RBC looks like a Cap with a thick rounded margin.This Cap shaped RBC is flexible just like the Cap.Due to this flexibility the RBCs are changing its shape when passing through smallest capillaries .These different shapes of the RBC is a normal phenomenon as they retain the original shape when they come out through these smallest capillaries (as these RBCs are now free from the pressure of the wall of the smallest capillaries ).Sometimes the change in shape is permanent.When central concave area is facing upwards the RBC looks like Ovate or Kidney shaped in light microscopy ,as the flattened peripheral part is not touching the surface of glass slide .But when central concave part is facing downwards , at that time the RBC looks like a Circle because this time the peripheral flat part is touching the surface of the glass slide.When the concave central part of RBC is facing sidewards it looks like Tear Drop or Cone or Helmet like (above conclusions were made after observing many light microscopic and the scan electron microscopic photographs available in the internet,and thus proving above observations
