It's probably just a fictional story from Uncharted 2. There is a documentary about the search of Shangri la. That talks about ancient text and cave dwellings in Tibet. There is some truth to the myth but I am not aware of any accounts of Marco Polo ever trying to find the lost city. Clearly Naughty Dog did there research though. Tibetan characters in the game are actually real Tibetan characters.
