On his exploration voyage of 1513, on which he named Florida, he had 3 ships and 200 men. He also rescued the crew of another Spanish ship that had followed him and was wrecked in a storm.

On his final voyage in 1521, he carried another company of about 200, but most were colonists for his proposed settlement on the Gulf coast in southwestern Florida. A native attack ruined his plans by chasing away the settlers, and poison from a native arrow eventually caused his death, shortly after their return to Cuba.
