Can't say this is all of them, but here's some:
1. Few if any civilized facilities:
-- paved roads
-- easy access to manufactured goods
-- access to repair for broken items
-- access to professional medical services
2. Unfamiliar territory (this has several implications):
-- sometimes no European folk had been through the territory yet, so no maps or experienced guides
-- didn't know what yearly weather patterns were like to know how bad the winter might be or how much or reliable the precipitation would be for farming
-- didn't know the quality of the soil for agriculture
-- didn't know which native plants were edible or useful for other purposes
3. Sometimes hostile natives
4. Sometimes hostile neighbors:
-- earlier prospectors/trappers didn't always appreciate settlers horning in on the land
-- some con men and robbers preyed on isolated settlers
There were rivers, dieseases, lack of food, rattlesnakes, and more.
