There is no difference exactly, but speaking to is more to do with YOU talking. Speaking with implies a group conversation where you are speaking AND listening. I think conversing is a better word for this, but that's just me :)

Of course the prayer of one person can affect a nation. We must remember that God would not talk to everyone. He always had a chosen one to tell the people what He wanted them to know. And, when the people were in trouble or had a problem, that chosen one would speak to God on their behalf.

Because you can't speak for someone you don't know. Example: I could ask you to speak in my behalf, i could tell you I'm a good person, but for all you know I'm a murderer that eats kittens for breakfast.
