Well, Mesopotamia, like all land on Earth, is far older than you can imagine, although I presume you actually mean when the civilizations IN the area were born. They were first born in the Neolithic Age (which began around 12,000 years ago), when hunter-gatherers settled down to lead mostly agricultural lives for the first time; over a period of many years, settlements would lead to villages, towns, cities, then city-states and empires. However, "real" civilization didn't begin until around only 5,000 years ago.
