You have to know the coin's denomination and date. Mint marks are in all sorts of different places, not even necessarily the same place during the same year!
One of the best places to look is a site such as www.coinfacts.com that tells you the location of every mint mark on every U.S. coin ever struck.
The first US Mint was located at t 7th and Arch Streets in Philadelphia. It opened in 1792. As demand for coinage grew, the Philadelphia Mint has occupied 3 other locations and is currently in city block-sized facility at 151 North Independence Mall just a short distance from the original building.

Mint mark on imperial era coins is located to the left and right of the eagle's tail on the reverse of the coin. At no point was it located above the denomination during this era. In any case, unless the coin is uncirculated, retail values are generally in the �0.50 to �2.00 range (maybe 65 cents to $2.50 U.S.) so this is coin does not command a large premium.

On an 1879 Silver Dollar, the mint mark is located on the reverse side, just below the eagle's tail feathers. Depending on the mint where it was produced, the mint mark will be either an "S" for San Francisco, "CC" for Carson City, or no mint mark for the Philadelphia mint. This small letter indicates the location where the coin was minted.
If it has one it would be located on the reverse side of the coin just above the letter "D" in the word "DOLLAR". The mint mark "S" is for San Francisco, California. The mint mark "CC" is for Carson City, Nevada. If there is no mint mark the coin was struck at the mint in Philadelphia, Pennsylvania.
The mintmark if it has one, is between the word ONE and the tip of the eagle's tail.
The mint mark on a 1976 $2 bill is located on the front side, in the lower right corner, just below the portrait of Thomas Jefferson. It appears as a small letter, either "E" for the Philadelphia Mint or "D" for the Denver Mint. If there is no mint mark, the bill was produced in Philadelphia.
