Rome's location or geography was significant because at its founding there were many warlike peoples surrounding them. Being located on and around hills, the city could be easily defended.

It was also located far enough inland to be safe from attacks by the sea, but yet have access to a seaport. Rome was also located near an easy crossing place on the Tiber, which gave the early settlers access to inland activities.
