3 pm , eastern time is when it ends . If you make a deposit after that , even though the bank doesnt close until 5, if you make the deposit after 3pm it counts as depositing in the next banking day

A banker usually works 6 to 7 hours but often the load of work being heavy, intricate and of high responsibility they work for more number of hours. It actually depends on the type of transactions taken place during the day which determines the load on a particular counter/table or desk.
