Two acceptable abbreviations for the plural of Ms. are Mses. or Mss. If you aren't comfortable with that format or the two females have different last names, you may address them individually: Ms. Mary Smith and Ms. Jane Jones; Dear Ms. Smith and Ms. Jones, Mss. If this is a personal letter to just this individual then you would address it: Mrs. Jane Doe or you could put Mrs. John Doe.Some women would find it offensive to call them by their husbands full name and just forgetting theirs altogether so address it Mrs. Jane Doe.
The address should read: Drs. John and Jane Doe or, Dr. John Doe and Dr. Jane Doe.

The proper way to address that letter would be "Mr. and Mrs. John Doe."
However you would address them individually with and in between, as:Mr. John Smith and Mrs. Jane Jones
If they are your friends then start the letter out with: Dear Jane; John and Joe. If you do not know the people well then start the letter with: Dear Jane Doe; John Doe and Joe Zero.
There are two ways to address a female doctor and her husband formally: * If it's a friendly get together then you could address them to each guest as 'This is Dr. Mary Jones and her husband Jack. * If it very formal then introduce them as Mr. Jack Jones and his wife Dr. Mary Jones.

I am fairly certain that you would address the letter by listing the names alphabetically on separate lines on the envelope, and then follow with the address. Example: Mr. John Jones Mrs. Jane Smith 200 Nowhere Lane Anyplace, State 00000-0000 It is handled differently if one of the individuals has a title (such as a doctor)...but, I can't remember the rule for that one.
