You would use Regards, not Regard in that situation.
The word 'regard' is used when you have finished the letter and want to sign it. Example: Regards, Ms. Jane Doe. In the text of of the letter another example: Dear Mr. Doe: In regard to your fax of October 16, 2010 (then finished the sentence.)
Warmest regards is an acceptable complimentary close for a friendly personal letter.
The term 'regards' is frequently used in letter-writing. 'Regards' usually forms part of the signature to a letter and is generally preceeded by an adjective, as in 'warm regards', 'fond regards', 'best regards', and so on. You are telling the person you're writing to that you hold them in good esteem, that you like and respect them. You might also use the term 'regards', 'regard' or 'regarding' in the body of your letter. You might say, 'I'm writing regarding your comments ...', or 'In regard to your question ...' In this instance you're simply telling someone what you're writing about. You could as easily say, 'I'm writing 'about' your letter ...' and so on.

To me "Best Regards" always sounds like some has mashed together "Best Wishes" and "Kind Regards". Of those two "Best Wishes" can be used when you have a close relationship with the recipient and "Kind Regards" seems appropriate for someone you have met before but still have some level of formality to the relationship.
When 'regards' is written at the end of a letter or email it is being used as a closing. The literal definition of 'regards' is to think of someone. The colloquial meaning of 'regards' as a closing is that the person writing the letter will keep you in their thoughts.
