Yes, but not as a result of enemy fire. While executing a 100 gun salute prior to their surrender, nearby ammunition was ignited by a spark and one member of the gun crew was killed. The other crewmen were wounded, and one of those later died of his wounds. The salute was reduced 50 shots, the flag was struck, and the Union troops withdrew.

I heard on a TV documentary that there was one casualty as a result of the battle; a Confederate horse.
No, noone died at The Battle of Fort Sumter. But a horse died.
