One method is to take one thread sample from the map and burn it. Rayon will melt but silk will burn.
Most "silk" maps made in WW2 were actually rayon. A few of the early maps were silk and these were usually crude maps and not in color. In 1943 the British also began using viscose rayon and cuprammonuium, a form of rayon commonly known asBemberg silk or copper rayon.
The following site identifies most of the escape maps used by the Americans and many of those used by the British.

Rayon was the first mandmade fiber to be produced and is known as a regenerated cellulose. Rayon is known as "artificial silk" and was created as a less expensive alternative to silk, although rayon is still an expensive fabric. It is easily wrinkles and shrinks, and is usually dry clean only. However, modifications can be made to rayon to make it stronger, known as HT Rayon. Modal is rayon that is washable.

Artificial silk, also known as rayon or viscose, is a man-made fiber produced from natural materials like wood pulp. Pure silk, on the other hand, is a natural fiber produced by silkworms. While both can have a silky texture, pure silk tends to be smoother, more luxurious, and more durable than artificial silk.
