The Hemalurgic power is what is used to create Inquisitors, Koloss and Kandra. With this power metal can be used to steal allomantic powers or human abilities (eg. strength, senses). If that metal becomes a part of your body it gives you that stolen power and, once removed, that power is gone.
The Koloss were once human until certain metal spikes are put into their bodies (mainly iron for strength). This also puts them under Ruin's control, as he can influence anyone with metal in their bodies.
There were 4 creatures that the Lord Ruler in the series. They were the Koloss, the inquisitors, the mistwraith and the Kandra. The Mistwraith and the Kandra are very close to being the same but the Kandra have a sentient mind.
