You subscribe to the trades to see if/when they're casting for James Bond, and then you go to the audition. This happens, roughly speaking, once a decade. James Bond has been played in "official" feature movies (not counting the 1967 Casino Royale, which was move of a pastiche/spoof and had no fewer than fiveBonds in the same movie) by:
* Sean Connery starting in 1962 (Dr. No)
* George Lazenby (one movie, 1969's On Her Majesty's Secret Service)
* Sean Connery again (1971, Diamonds are Forever)
* Roger Moore starting in 1973 (Live and Let Die)
* Timothy Dalton starting in 1987 (The Living Daylights)
* Pierce Brosnan starting in 1995 (Goldeneye)
* Daniel Craig starting in 2006 (Casino Royaleremake)
Daniel Craig is scheduled for the next two movies, but it's possible that they'll need a replacement after those wrap, as his ten years will be about up by then. He could hold on a bit longer (Roger Moore was Bond for almost 15, and Connery did Never Say Never Again in 1983, a full two decades after first appearing in the role ... it wasn't an "official" Bond film, though).
There are also Bond pastiches/parodies. Your best bet there is to look and sound like one of the guys from the feature films, preferably either 1960s Connery (as the most iconic) or whoever's currently doing them (as the most recent).
The main thing to remember about James Bond is that he is not a womaniser. Ian Flemming's original character suffered abandonment issues, which is why he frequently has intercourse with numerous women, and does not pursue a longer relationship.
Besides that, its all about being suave. Bond, as a character, is cool, calculating, and never gives away more than he needs to. The character never deliberately makes a scene, or hits on women, unless he needs to. Its all about being cool, and not over doing it. Suits are not required, but a decent sense of style will help. Bond is never seen in jeans, but on occasion has wore hawaiian shirts, with dress pants.
