It is never (to my knowledge) blatantly spelled out in the anime or movies that Misty loves Ash or vice versa, but it is definitely implied. Throughout the first five seasons of the anime, there are hints put in that could suggest that she does, the greatest probably being "The Heartbreak of Brock." Ash says "Imagine Brock being married" in which Misty responds to Ash saying "You and I will be married some day." Also, here is dialogue from that episode while Misty is talking to Brock:
Misty: Take it from me, it's a lot easier to like someone who likes you than to like someone who doesn't.
Brock: How do you know?
Misty: (blushing) Well, that's what I've heard.
Also, in "Gotta Catch Ya Later," Misty gets upset when she thinks Ash is taking her for granted by saying now that her bike is fixed, she can get to Cerulean City faster. She even cries over it, and right before they part ways, she even sings a SONG about Ash, talking about how he's more than just her best friend.
There are countless other hints like this. The writers of the anime most likely put in underlying romantic suggestions but don't blatantly spell them out to keep people more hooked to the shows (and movies). They do the same with May and Dawn (but not the extent they did with Misty)... Just look on YouTube: There are COUNTLESS Ash+Misty, Ash+May, and Ash+Dawn fan-made videos, pictures, slideshows, etc... And ultimately this helps the writers of Pokemon keep their viewers more interested in the shows and the franchise.
But as I said, it is never blatantly spelled out (to my knowledge) that Misty (or May or Dawn for that matter) loves Ash, or vice versa (although it's implied the most with Misty in the first few seasons). Mostly, we fans seem to want to believe otherwise hence all those fan-made projects, and those fan-made projects probably affect how we view Ash's interactions with the various female companions he meet.
