John Glover did not leave Smallville of his own choice the writers of the Smallville series had made the decision to write him out by making it so that his own son; Lex Luthor would murder him however the writers later brought John Glover back in Season 10 as Lionel Luthor but as Lionel Luthor from another alternate reality in which the Luthors were the one who had found and adopted Clark Kent and therefore became Clark Luthor.
Ian Somerhalder's character in "Smallville" was named Adam Knight but he was once known as Chad Nash. Chad died of cancer but Lionel Luthor had brought him back to life with the Lazarus serum which was serum that was created using Clark Kent's blood. Adam dies because Lionel no longer would supply the serum to Doctor Teng.

Smallville had wrote Sam Jones III's Pete character out during Season 3 because they had no clue what to do with him so they wrote him out by explaining that he had moved with his family to another city in Kansas however Pete returns in "Hero" which is the 7th season's 13th episode just to visit Clark and his friends for the episode and while he's in Smallville he's been infected by Kryptonite gum and he also learns from Clark that Chloe Sullivan, Lionel Luthor and Lana Lang all had learned about Clark's Kryptonian secret while he (Pete) was away.

No, Ian Somerhalder and Paul Wesley no longer work on "Smallville." Ian Somerhalder had played a Season 3 character named Adam Knight (previously known as Chad Knight.) Adam Knight was a character that had been dead but Lionel Luthor had resurrected him with an experimental drug and had him spy on Clark Kent in an attempt to find out what he was hiding. Adam ended up dying for good in the episode titled "Crisis." Paul Wesley only appeared in the Season 2 episode titled "Prodigal" as Lucas Lothor, the lost long brother that Lex Luthor had.
