It's kind of hard to be more specific than that without giving you an entire theatre course, but basically they look for things that don't mesh with their concept of the production and correct the actors when necessary. They may also have actors try multiple blockings or readings of a scene to see which "plays best."
The first time a particular scene is rehearsed, they'll also give the actors their initial blocking. This may be vague ("By the time you're done speaking you need to be standing right behind her") or more detailed ("On this line, cross up stage right to here, then turn when she says this line, then move directly toward her slowly throughout your next speech until you're standing right behind her at the end of it").
