They FIRST Kissed under a magical spell in "Something Blue" when they thought they were in love and going to get married. Later She also (not under ANY magical influence) gives him a short sweet meaningful kiss for not telling Glory about Dawn being the key and taking a very bad beating while not giving up who the Key was also I have to add. THEN a very long passionate kiss as another person said below at the end of the musical episode. (but she does say/sing right before kissing him at the end of - Once More With Feeling- that "this isn't real but I just Want To Feel") so it is misleading as to whether she means it or not, but as any Buffy & Spike fan knows, she already had feelings for him at this point.
