Yes, the famous Scarlet and Rhett staircase scene did take place at the Jefferson Hotel in Richmond, VA. I happened to stay at the Jefferson Hotel in VA a number of years back. The hotel was recommend by a local of the area and I was faithfully informed that filming was taken at the location. What did I do when I saw the staircase? Ran up and down and up and down the staircase calling Rhett. He didn't answer (smiles). Incidentally, although the staircase beautiful in craftsmanship, isn't that big. UPDATED ANSWER!!!
