After an interaction with Prince Fiyero, Elphaba sings 'I'm Not That Girl'. She's concerned about the rights and well being of animals in Oz. Of particular concern are those animals that end up caged. Caged animals don't learn to speak. It's the ability to speak that differentiates Animals from caged animals.
As a student at Shiz University, Elphaba finds a caged lion cub in history class. She casts a spell, and gets the cub out of the classroom with Fiyero's help. The two set the future Cowardly Lion free in the woods. Fiyero quickly leaves when he realizes that he could fall in love with this green skinned nonroyal. It begins to rain. Elphaba seeks shelter under a bridge. During the storm, she thinks of the impossibilities of a wealthy young prince falling in love with her, and starts singing 'I'm Not That Girl'.
