The use of valves changes the length of the tubing and therefore its resonant frequency. Tuning of the instrument is done using either electronic tuners or mechanical tuning forks as a guide to correct pitch then the tuning slide is extended or contracted to set the base pitch. However, its not often for brass instruments to need tuning. They are made in the the key of C as a majority, but have B-flat versions as an alternative. The tuning slide is normally set using the main slide or tube which comes from the mouth piece to the 3rd valve in the first instance. There are 2 other slides that are used that is first valve and 3 valve slide. C-sharp normally requires the 3rd valve extended even if the trumpet is concert tuned.

What does 528hz sound like?
This is a good example of an actual 528 Hz tone: See related link to youtube for the example528Hz is a C (actually closer to a C#) in the western musical scale. Instrumental tuning imposed mid-century is set at the note A being 440Hz. If a musician tunes their instrument using the 528 Hz frequency, the A note should be 444 Hz. You can try this at home. There are many tuners and explanations online. Resonating this forks is also oddly very fulfilling.
How do you tune a cello right?
It depends on the cello, and how far it is out of tune. You can use an electronic tuner (one with a little needle to show how far out of tune) or you can use an electronic tuning tone generator, a tuning fork, pitch pipe or even a piano if you have one. Some beginner cellos have fine tuners on all four strings (these are the little screws sticking out of the triangular piece towards the bottom) all cellos have pegs which offer a wider (less precise) method of tuning, which should only be touched by someone experienced. You should leave it to a teacher to tune your cello correctly, and have them show you how, as it is very difficult to show you how over the internet.
