The lub sound occurs around the peak of the R wave in an ECG because it is associated with closure of the mitral and tricuspid valves, which happens at the beginning of ventricular contraction. This coincides with the R wave, which represents ventricular depolarization and the onset of ventricular systole.
The second sound of the cardiac cycle, commonly referred to as the S2 sound, occurs when the aortic and pulmonary valves close at the end of ventricular systole as blood is forced out of the ventricles and into the aorta and pulmonary artery. This sound marks the end of the systole phase and the beginning of diastole.
Ventricle diastole is the period during which the ventricles are relaxing. During ventricular diastole, the pressure in the (left and right) ventricles drops from the peak that it reaches in systole. When the pressure in the left ventricle drops to below the pressure in the left atrium, the mitral(bicuspid) valve opens, causing accumulated blood from the atrium to flow into the ventricles.
