Photosynthesis has two stages.

1: Light reaction / Light dependent reaction

2: Dark reaction / Light independent reaction

Light reaction requires light whereas dark reaction does not require light however it also occurs during the day.

Light reaction :

Light reaction occurs in grana (thylakoid stack) of chloroplast. Thylakoid membrane encloses space called lumen. Outside the thylakoid is matrix called stroma. Protein complexes in the thylakoid membrane for absorption of light are Photosystem. Photosystem l (or plastocyanin) absorbs light of wavelength 700nm. Photosystem ll (or water-plastoquinone oxidoreductase) absorbs light of wavelength 680nm.

During light reaction, photosystem ll absorbs light and its electron get excited and leave the photosystem ll . It regains the electron from water molecule and splits it into O2 and H+.

Plastoquinone gains two H+ (protons) from stroma and two electrons from photosystem ll. It transports protons into the lumen while the electrons move towards the cytochrome b6f protein complex.

Cytochrome b6f transfers electron from plastoquinone to plastocyanin. Then photosystem l (P700) accepts electrons from reduced plastocyanin. Ferredoxin is soluble protein complex in photosystem l that carries electron from iron-sulfur complex and transfers it to ferredoxin-NADP+ reductase. Ferredoxin-NADP+ reductase reduses NADP+ to NADPH.

H+ which were pumped into lumen through plastoquinone go back into the stroma by passing through ATP synthase. As H+ ions pass through ATP synthase, they cause it to spin. Phosphate group and ADP on it come closer to form ATP.

Therefore the products of light reaction are NADPH and ATP which are later used in the light independent reaction.

Dark reaction / Light independent reaction:

It is also called calvin cycle. During dark reaction, 6 CO2 molecules bind with 6 ribulose biphosphate (RuBP) which contains 5 carbon atoms and two phosphate groups. Together they form 12 molecules of PGAL (phosphoglyceraldehyde ) which contains 3 carbon atoms and one phosphate group (3 C atoms in each PGAL=36 C atoms; 6 carbons in six molecules of CO2 and 30 carbon atoms in six 5-C molecule RuBP= 36 carbon atom). During conversion of six CO2 molecules and six RuBP into 12 PGAL molecules, 12 ATPs and NADPH (produced in light rection) are converted into 12 ADPs, phosphate group and NADP+ in order to provide energy for dark reaction. Enzyme that catalyses this reaction is RuBisCo (ribulose biphosphate carboxylase).

Out of these 12 PGAL molecules, 10 are used to regenerate RuBP (ten 3-C molecules = 30 carbon atoms; six molecule of 5-C RuBP = 30 carbon atoms). In this reaction 6 ATPs are converted into 6 ADPs and phosphate groups. Therefore ten PGAL were required to regenerate six molecules of RuBP whereas 2 molecules of PGAL are used to make a glucose molecule. Glucose is a six carbon molecule; 2 PGAL molecule also contain six carbons (3 in each).

Now the overall reaction in the process of photosynthesis:

6CO2 + 12H2O + sunlight ---------> C6H12O6 + 6O2 + 6H2O
