When you are doing an ELISA you need to have a specific antibody to target your protein and it can be difficult to have access to the antibody that you want.

You can also have problems doing the washes:

- When you wash your plate after adding the antibodies, some of them (that are not linked) can remain there which will affect your results. So you can have false positive results.

- You can wash away some proteins accidentally (including the one that you want identify). So you can have false negative results.

One limitation of ELISA is that it can sometimes have issues with specificity, leading to false positive results. Additionally, ELISA may not be as sensitive as some other detection methods for low levels of analytes. Timing and manual steps involved in the assay can also introduce variability.
