Most of the times, when a mass of E. coli or eukaryote cell mass is being collected from a vessel (e.g., a bioreactor), the cell mass, either intact or disrupted (lysed) cells, is kept on ice to prevent to activate nucleases that can chew the target nucleic acids. Considering that during extraction methods, a large number of the cells die, it is important to prevent the enzymatic activity begins during cell death process.
