All Japanese vowels are always pronounced the same way, although when next to other vowels and/or spoken quickly, it can sound like there are different pronunciations.

"Kudasai" is pronounced "koo-dah-si." The first syllable is a hard k followed by a short o. The second syllable is pronounced like the second syllable in "panda" and the last is simply an s followed by a long i.
