In Act 5 Scene 9, Macduff got Macbeth to fight by calling him the names 'hell-hound' and 'bloodier villain'. At first, Macbeth wasn't worried by the witches' warning to fear Macduff or the movement of Birnam Wood to Dunsinane. Instead, he chose to concentrate on the cautionary advice that seemed incomprehensible and impossible to him: defeat at the hands of a man that hadn't been born of woman. So he figured that he just needed to watch out for any tricks that Macduff might be up to. He didn't realize that he needed to watch out for his life. In essence, Macduff was a man who hadn't been born of a woman. He had been born by a Caesarian delivery from a dead mother. But the realization that the fight that already was in progress was doomed didn't stop Macbeth. As he always had done on all the battlefields of his life, Macbeth fought bravely to the death, be it his or his opponent's. In this case, his luck finally had run out.

Macbeth didn't want to fight Macduff, because he didn't want to add any more bloodied casualties to the Fife Castle massacre body count. In Act 4 Scene 2, Macbeth [c. 1014-August 15, 1057] had Macduff's entire family and household massacred at the Macduffs' home of Fife Castle. In Act 5 Scene 8, Macduff finally caught up with Macbeth and challenged him to fight. Macbeth didn't want to fight for two reasons. The reason that he didn't reveal was the witches' warning to beware of Macduff. The reason that he did reveal was the weight on his soul from having ordered the Fife Castle massacre.
Macbeth and Macduff got into a fight because Macduff sought to avenge the murder of his family, which Macbeth ordered in his ruthless quest to secure his throne. Macduff held Macbeth responsible for the devastation and sought to bring him to justice.
Lady Macbeth does not convince Macbeth to kill Macduffs family. She convinces him to kill the King at the start of the play, but after that she has no input in the decisions he makes. Macbeth chooses to have Macduffs family killed so that he may seize the Macduff castle for his own. The witches also tell him to 'beware Macduff' when he visits them and so he feels threatened by him. Macbeth doesn't actually personally kill Macduffs family either, he hires the same thugs that he hires to assassinate Banquo and Fleance to murder him.
Malcolm initially mistrusts Macduff because of his close association with Macbeth. However, he later recognizes Macduff's loyalty and bravery, and comes to see him as a valuable ally in the fight against Macbeth.
