Shakespeare did not write down any of his feelings about anything, so it is hard to know. It is hard to imagine that he was not saddened by the event, but it is also far too easy to project our own feelings onto him. Infant mortality was much more common back then and perhaps was not always as devastating as it is to people nowadays.
It is easy to find characters in Shakespeare's plays who are fathers grieving for dead children, and even easier to say "Well, that was Shakespeare grieving for Hamnet". The problem is some of them are from plays written before Hamnet died (like Henry VI Part III) and others from plays written many years after (like Macbeth).
Bottom line: never ask about how Shakespeare felt about anything, because you can never get any kind of reliable answer.
