"Fust" means "become dessicated or withered". The snarky character Thersites says in Troilus and Cressida " if he knock out either of your brains: a' were as good crack a fusty nut with no kernel." We've all met those nuts--you crack them open and there's only a withered slip instead of a proper kernel. Well, Hamlet says that if we do not exercise our capability and godlike reason, it will dry out and wither from lack of use. And when he says "Sure he that made us with such large discourse, Looking before and after, gave us not That capability and godlike reason To fust in us unus'd." he means that God did not give us our brains to dry out from lack of use.
