The three witches predict that Macbeth will be king, so he kills King Duncan in order to make that happen. However, the witches also predict that Banquo will produce a line of kings. Macbeth wants Banquo and Fleance (Banquo's son) killed so that they cannot take the throne away from him. However, his plan backfires when Fleance escapes.

Macbeth wanted Banquo and fleance dead because they are a threat to this position as king knowing that the the witches have predicted that Banquo will produce a line of Kings. This is also a sign of Macbeth's paranoia.

How did the murders kill fleance in the play Macbeth?

In the witch's prophecy, Banquo is told that he will not be a king, but his sons will. This means that Macbeth will be king, but something will happen which prevents his bloodline from maintaining the throne, which will be taken by the bloodline of Banquo. In Macbeth's paranoia about maintaining the throne, he orders Banquo and his son killed, cutting off their bloodline and keeping them from usurping the throne from Macbeth. However, Fleance escapes, and while Malcolm takes the crown, it can be inferred that Fleance or one of his descendants take it from Malcolm or one of the later Kings.

Did Lady Macbeth want Macbeth to kill Banquo?

Lady Macbeth wasn't in on her husband's plans to kill Banquo. When she asked what his plans 'of dreadful note' [Act 3 Scene 2 Line 44] were, he didn't want her to know the specifics. But most likely yes, she would have wanted Macbeth to kill Banquo. Earlier in their conversation, she commented that Banquo and his son Fleance were mortal [and therefore killable?].

Why does Macbeth want to know whether or not Fleance is going riding with his father in the afternoon before the banquet?

Macbeth wants to know wether or not fleance is going riding with his father in the afternoon before the banquet because if they are riding together it will be easier for the murderers to kill both of them and "kill two birds with one stone."

What does the conversation with the murderers reveal about Macbeth?

The conversation he had with the murderers show that he is still trying to cloak his crime under a veneer of political necessity, by suggesting that the murderers have good reason to hate Banquo and wish him dead. In reality they are hired murderers, who would kill anyone if they were paid to do it. They are quite happy to attempt to kill Fleance, who is obviously not their enemy. So, what is Macbeth on about? He is trying to justify the murder to himself, because he still has enough conscience to think that it is wrong to kill his friend just because his decendants might become kings someday.
