Within the context of the play, the letter didn't get to Romeo because Friar John, who was supposed to be carrying it, got himself quarantined because he was suspected of having the plague.
Shakespeare has probably done this so that the story is more dramatic. The story probably wouldn't have been half as good if he received the letter, and nor would it have had the same outcome, so it probably wouldn't be labeled a tragedy.
Sadly, the message was never delivered to Romeo, there was a mix up in the mail or something like that. It's like today, when a text message never goes through. Sadly, that leads to their deaths realizing neither can live without each other.
