The ark contained the Two "Tablets of Testimony" (so called in Exodus 31:18) on which the ten commandments were written. The source is Exodus 25:16 "You shall place in the Ark the Testimonial-tablets that I shall give you."
No one knows for sure. There is some speculation that it is under Golgotha Hill because of Ron Wyatt's testimony. However, the last mention in history of it's whereabouts is in 2ND Maccabees which states that Jeremiah took it and the Tent of Meeting to Mount Nebo and hid them in a cave there. Most certainly, the Ark was not mentioned in service past the beginning of the Babylonian captivity.
