When Adam and Eve sinned, their eyes were opened to their naked state. It's been said that they had been formally covered by glory in their innocence and that covering was lost. Realizing they were naked, they sought to cover themselves, and so used fig leaves to try to create clothing. While the act of sin caused the immediate beginning of death in their bodies (what we think of as aging), the concept of death was still unknown to them. Moreover, while they knew they had done wrong, they did not know how it would need to be made right. God had to kill animals to create clothing in symbolism of the fact that blood is needed to pay for sin. This is a foreshadowing of Christ's blood on the cross covering all of man's sin. One common interpretation is that this is a symbol of man trying to cover his sins himself. It doesn't work long-term. God had to step in.
