There is no specific room in a Church for groomsmen, you would have to ask the priest in your particular Church. I have seen groomsmen wait in the vestibule, the Vestry, the Sacristy, the narthex, and the porch (having a cigarette); they could also wait in the cryroom, the social hall, the basement, well, practically anywhere.
