The "Colonial" Puritans failed to convert the Indians because ultimately, they failed. Their harsh intolerance, along with other poor decision making (Salem Witch Trials) led to their own destruction.

Tobacco was successful because of the profit it made. It could be sold for a good price to England and the colonists could grow plenty of it due to Jamestown's location (Virginia which has a great climate for growing crops.
If you did not work in Jamestown, then you did not eat. The colonists planted food and built shelters and fence. They made peace with nearby Indians.
