You cannot play as Reptile in Mortal Kombat 1. You can only fight against him. Although there are glitches, where if Player 2 presses start to enter a fight while Player 1 is fighting Reptile, the Ninjas will appear as Reptile but will not fight like Reptile. There isn't even a Game Genie code to fight as him. The Dragon symbol in the middle of the character selection screen does not ever change into another character.
This answer is half right and half wrong. In the snes version of the game after defeating shang tsung he will free all the souls he has captured. when you see sub zero the first time he appears, hold L and R (The trigger buttons) don't let go of the buttons until the title screen appears and then start a new game. you will see reptile's face in the upper middle of the screen. enjoy
