Significant means important, or outstanding; the term doesn't suggest whether significance indicates a good thing. For example:

Mother Teresa made a significant contribution to the health and well-being of people suffering from poverty.

Mother Teresa fearlessly tackled Calcutta's significant problems with poverty, illness and deprivation.

Smallpox was a disease which caused significant suffering and death.

Dr Edward Jenner made significant discoveries in the fight against smallpox, which led to its being eradicated.
