I have visited a couple of contenders for this prize:

1. Santa Sophia in Istanbul which has been church, mosque and museum since it was completed in 537 AD.

2. The Pantheon in Rome which was built as a temple by Hadrian in 117 AD, if I remember correctly, and is also now a museum.

I would be very interested to hear of other old buildings which have been in continuous use for over 1500 years.

Additional commentsPer Wikipedia, as of 2009, the Pantheon is still in use as a church. (But Santa Sophia is a museum). The Catholic church Santa Sabina in Rome, built in 422 CE (AD), is substantially the original structure and is still in use. There are some other churches originally built in the 4th century CE still in use, but these churches have been substantially reworked, and I am not sure how much of the original buildings survive. In addition, the Mausoleum of Hadrian, completed in 139 CE, and converted to a fortress around 400 CE, is still in use as a museum.
