Well, it depends on where you're coming from. But generally speaking, you will need to take the 4-5-6 (the green line, aka the IRT or the Lexington Avenue line). The 4-5-6 is currently the only subway line that goes up and down the East Side.

You could also take the N, R, or W (of the N-R-Q-W, the yellow line) to Lexington Avenue-59th Street. Lexington Avenue is three blocks east of Fifth Avenue.

Or, you could take the E (of the A-C-E, the blue line) or the V (of the B-D-F-V, the orange line) to East 51st Street-Lexington Avenue. That is the last stop on the Uptown E or the V before they go to Queens (and the first stop on the Downtown E or V fromQueens).
