If you take the highway, the drive from Portland to Bar Harbor is about four and a half hours - give or take. Now if you decide to take the scenic route, which is highly recommended, plan on driving a few more hours. Of course the time of year also has an impact depending on traffic and tourists. For a short cut and some local scenery, Exit to Rt 3 near August go East through Belfast and Bucksport to Ellsworth, then on to Bar Harbor a few more minutes down the road. Belfast has a Maine-looking harbor and old timey main street. Near Bucksport is the new bridge over the Penobscot river. It has an observation deck inside one of the support towers well worth the visit.
