Rio Grande
At the outbreak of the Mexican-American War in 1846, the province claimed by the United States was Texas. Texas had declared independence from Mexico in 1836 and was annexed by the U.S. in 1845, which heightened tensions between the two nations. The dispute over the border, particularly the claim that the Rio Grande was the southern boundary of Texas, further escalated the conflict.

Texas itself; before the Texas Revolution (1835-1836) Texas had been a territory of Mexico and until the Mexican-American War (1846-1848) it was seen as a rebel province to acquire on a later date.

No. The Alamo was a battle in the Texas Revolution, where Anglo settlers of Texas (most from America) were rebelling against Mexico. Texas at that time, 1836, still belonged to Mexico. The Texans eventually won that war and became independent of Mexico. Texas was an independent nation for about ten years. Then the Texans decided to join the US. A dispute over the southern boundary of Texas brought on the US-Mexican War of 1846-48. Lee did fight in this later war.
