The Manhattan spoke an Algonquian language - part of a very large language family. They probably spoke a language related to Wappinger, or a Lenape (Delaware) dialect. These two are very closely related languages.

The colonists that learned how to speak native languages were usually the children. They would exchange boys to learn each other's language, then the boys would become interpreters. There were also those people that were captured or married into the tribe/colony.
